Development and validation of stability indicating HPLC–PDA method for the simultaneous determination of benzoyl peroxide and tretinoin in topical dosage form

Abstract Background Acne vulgaris is a very dangerous skin disease leads to psychological disorders. Benzoyl peroxide and tretinoin in combination with topical dosage are used improve the appearance of acne-prone skin. The study aimed develop validate an HPLC–PDA method for simultaneous estimation both drugs. Intentional modifications were implemented analytical get better optimum conditions. final was chosen reverse phase chromatographic separation by using C18 column as stationary 0.01 M phosphate buffer adjusted pH 2.5 mixed acetonitrile (25:75 v/v) mobile phase. optimized conditions 1.5 mL/min flow rate, 30 °C temperature, 5 autosampler 20 µL injection volume. wavelength detection at 272 nm Tretinoin 353 utilizing PDA detector. All standard sample solutions made methanol. Results developed exhibited peak retention times 2.94 min 11.34 Tretinoin. This proven be robust, linear calibration curve, accurate, precise, specific. forced degradation studies results showed that high degree specificity obtained separating analytes from produced impurities completely. Conclusions fast, economic stability indicating. It useful routine pharmaceutical analysis where benzoyl formulated their quality safety.
